พาวเวอร์พัฟฟ์เกิลส์ เกิดจากการทดลองที่ผิดพลาดของ ศจ.ยูโทเนียม โดย ศจ.ยูโทเนียม ต้องการเด็กหญิงสมบูรณ์แบบ โดยการผสมน้ำตาล เครื่องเทศ สารพัดของกุ๊กกิ๊ก แต่ว่าเติมสารเคมีX โดยไม่ได้ตั้งใจ แล้วเกิดเป็นเด็กหญิง 3 คนได้แก่ บลอสซัม บัตเตอร์คัพ และ บับเบิลส์ พวกเธอเกิดมาพร้อมกับพลังพิเศษ และใช้พลังนี้ในฐานะซูเปอร์ฮีโร่เพื่อช่วยปกป้องโลก ส่วน ศจ.ยูโทเนียม เขาก็รับหน้าที่เลี้ยงดูเหมือนกับเป็นพ่อคนหนึ่ง และคอยให้คำแนะนำสั่งสอน 3 สาวเป็นประจำ ทั้งยังคอยประดิษฐ์คิดค้นของใหม่ ๆ ให้กับพาวเวอร์พัฟฟ์เกิลส์อีกด้วย
Professor Utonium is hosting a party for all the rich and famous people in Townsville, so Bubbles makes sure that she is the most beautiful of the girls by continually grooming herself. While doing this, she notices one of Octi's legs poking out from under the girls' bed, so she goes to pick him up to find that all that's there is one of Octi's legs! After waking up from the shock, she vows to exact revenge on the person who 'killed' Octi.
